Abstract:

The aim of this study was to determine the quality of cumulus-oocyte-complexes (COCs) and the effects of bovine serum albumin (BSA) supplementation on in vitro maturation and fertilization rate of buffalo oocytes. COCs were collected from slaughterhouse buffalo ovaries by aspiration method. Only normal grades COCs were matured for 48 hours in TCM-199 media. Two groups were created: one for the maturation medium supplemented with 5% of BSA, the other without supplementation (control). Matured oocyte fertilized with capacitated frozen-thawed semen in Brackett and Oliphant (BO) medium for 5 hours, in an incubator at 38.5 °C with 5% CO2 underhumidified air. A significantly higher number of normal quality COCs per ovary (P

Effects of supplementation of the maturation media with insulin on in vitro maturation and in vitro fertilization of bovine oocytes.

This study was carried out to determine the effects of supplementation of the maturation media with insulin on in vitro maturation and fertilization of bovine oocytes. In Experiment 1, cumulus-intact bovine oocytes were cultured in a maturation medium (TCM-199 containing 10% fetal calf serum, 0.02 U/ml follicular stimulating hormone and 1 microgram/ml estradiol-17 beta) with or without insulin ...

Effects of fetal bovine serum and estrus buffalo serum on maturation of buffalo (Bubalus bubalis) oocytes in vitro

AIM The aim was to assess the effects of fetal bovine serum (FBS) and estrus buffalo serum (EBS) on in vitro maturation rate of oocytes in buffalo. MATERIALS AND METHODS Maturation rate of oocytes was assessed in two maturation media supplemented with 20% FBS and EBS. Oocytes maturation rate was evaluated on the basis of cumulus cell expansion and extrusion of polar body after 24 h of in vitr...

Post Thaw Recovery of Buffalo Oocytes Vitrified in Propylene Glycol and Trehalose supplemented with Bovine Serum Albumin

The present study has been undertaken to assess the post thaw recovery of buffalo oocytes vitrified at different stages of in vitro maturation (IVM) by propylene glycol and trehalose supplemented with bovine serum albumin (BSA). Cumulus oocyte complexes (COCs) obtained from slaughterhouse ovaries were randomly divided into 6 different groups: control (nonvitrified oocytes were matured for 24 h ...
